MOTS-C – often written MOTS-c, short for 'mitochondrial open reading frame of the 12S rRNA type-c' – is a 16-amino-acid peptide encoded in the mitochondrial genome rather than in nuclear DNA. First described in 2015, it belongs to the family of mitochondrial-derived peptides (MDPs), alongside humanin.

The MOTS-C sequence lies within the 12S rRNA region of mitochondrial DNA. In cell and animal models it has been linked to AMPK signalling – one of the cell's central energy sensors – and to the regulation of glucose and fatty-acid metabolism. In cultured cells under metabolic stress, MOTS-C has also been reported to move into the nucleus, which makes it a subject of research into communication between mitochondria and the nucleus.
MOTS-C is studied in models of metabolic homeostasis, insulin sensitivity, skeletal-muscle metabolism and exercise physiology, as well as in ageing research. As one of the few known peptides encoded by mitochondrial DNA, it is also of interest to groups working on mitochondrial genetics and mitochondria-to-nucleus signalling. All of this is research in cell and animal models.
Store the lyophilised vial at −20 °C, dry and protected from light; 2–8 °C is fine for short periods. Reconstitute with bacteriostatic water, added slowly along the inside of the vial, and swirl gently until dissolved – don't shake. Keep the solution at 2–8 °C, use it within about four weeks and avoid repeated freeze–thaw cycles.
