What is Kisspeptin?

Kisspeptins are a family of peptides encoded by the KISS1 gene. The gene was first described in cancer research, which is why the longest form was originally named metastin. Today kisspeptin is mainly known as a key upstream regulator of the reproductive axis.

Virexa Labs supplies Kisspeptin-10, the shortest biologically active form, as a lyophilised research peptide. It is intended for in-vitro laboratory research only and is not for human or veterinary use.

Structure and mechanism

The KISS1 gene produces a precursor protein that is cleaved into several kisspeptins of different lengths, such as kisspeptin-54, -14, -13 and -10. They all share the same C-terminal sequence, which ends in an amidated Arg-Phe motif (RF-amide). Kisspeptin-10 consists of exactly these final ten amino acids – the minimal sequence needed to activate the receptor.

Kisspeptin acts on the KISS1 receptor (KISS1R), also known as GPR54, a G-protein-coupled receptor. In the hypothalamus this receptor is found on GnRH neurons. Activation of KISS1R is studied as an upstream step in the release of gonadotropin-releasing hormone (GnRH), which in turn regulates the release of LH and FSH from the pituitary – which is why kisspeptin is described as a gatekeeper of the hypothalamic-pituitary-gonadal (HPG) axis.

Store lyophilised Kisspeptin at −20 °C, dry and protected from light (2–8 °C short term). For reconstitution, bacteriostatic water is usually used: add it slowly along the vial wall and swirl gently until dissolved – do not shake. Keep the solution at 2–8 °C, use within about 4 weeks and avoid freeze–thaw cycles.
