What is Tesamorelin?

Tesamorelin is a synthetic analogue of growth-hormone-releasing hormone (GHRH), the hypothalamic peptide that signals the anterior pituitary to release growth hormone. It is supplied by Virexa Labs as a lyophilised research peptide for in-vitro laboratory research only – it is not intended for human or veterinary use.

Because Tesamorelin is built on the full-length GHRH sequence rather than a shortened fragment, it is a useful reference compound in studies that compare different GHRH analogues and their behaviour at the GHRH receptor.

Structure and mechanism

Tesamorelin comprises the complete 44-amino-acid sequence of human GHRH – GHRH(1-44) – with one key modification: a trans-3-hexenoyl group attached to the N-terminal tyrosine. Native GHRH is rapidly cleaved at its N-terminus by the enzyme dipeptidyl peptidase-4 (DPP-4). The hexenoyl group was introduced to make this end of the molecule more resistant to that enzymatic breakdown, while leaving the receptor-binding sequence intact.

Mechanistically, Tesamorelin acts as an agonist at the GHRH receptor (GHRH-R), a G-protein-coupled receptor on pituitary somatotroph cells. Receptor activation triggers the cAMP signalling cascade that is studied as the upstream step of GH synthesis and release.

Store lyophilised Tesamorelin at −20 °C, dry and protected from light; 2–8 °C is acceptable for short periods. For reconstitution, bacteriostatic water is usually used: add it slowly along the inside wall of the vial and swirl gently until dissolved – do not shake. Keep the solution at 2–8 °C, use it within about 4 weeks and avoid repeated freeze–thaw cycles.
