What is SNAP-8 (acetyl octapeptide-3)?

SNAP-8 is the common name of acetyl octapeptide-3, a synthetic peptide of eight amino acids with an acetylated N-terminus and an amidated C-terminus. It is an elongated analogue of acetyl hexapeptide-3: the hexapeptide sequence is extended by two amino acids, giving Ac-Glu-Glu-Met-Gln-Arg-Arg-Ala-Asp-NH2. Acetylation and amidation remove the charged ends of the chain – a common modification that tends to make short synthetic peptides more resistant to breakdown by exopeptidases in experimental systems.

The name points to SNAP-25, the protein whose N-terminal region served as the template for this family of peptides.

The mechanism studied: the SNARE complex

The SNARE complex is a protein assembly that drives the fusion of vesicles with the cell membrane – a key step in the release of neurotransmitters such as acetylcholine at the neuromuscular junction. Its core components are SNAP-25, syntaxin and synaptobrevin (VAMP).

Because SNAP-8 mimics part of SNAP-25, it is studied as a peptide that may compete with SNAP-25 for its position in the complex and thereby modulate how the complex assembles. This makes SNAP-8 a tool compound in neuromuscular and cosmetic research models that examine vesicle release, mostly in vitro and often in direct comparison with acetyl hexapeptide-3.

SNAP-8 is water-soluble and is usually reconstituted with bacteriostatic or sterile water. Add the solvent slowly along the inside wall of the vial and swirl gently until the powder has dissolved; avoid vigorous shaking. Let the vial reach room temperature before opening, so that no moisture condenses on the powder.
