About SS-31

What is SS-31 (elamipretide)?

SS-31, also known by its non-proprietary name elamipretide and the code MTP-131, is a synthetic, mitochondria-targeted tetrapeptide. It belongs to the Szeto-Schiller (SS) peptides, named after the researchers who developed them. Its sequence is D-Arg-Dmt-Lys-Phe-NH2, where Dmt stands for 2′,6′-dimethyltyrosine.

Structure and mechanism: binding to cardiolipin

SS-31 has an alternating aromatic–cationic structure and carries a net positive charge. This allows it to cross cell membranes and concentrate in the inner mitochondrial membrane, where it binds cardiolipin – a phospholipid found almost exclusively there and important for organising the electron transport chain. The D-arginine at the N-terminus and the amidated C-terminus make the peptide more resistant to enzymatic breakdown.

Research areas

SS-31 is investigated in models of mitochondrial dysfunction, oxidative stress, ATP production and mitochondrial membrane integrity. Research groups use it in cardiac, skeletal-muscle, renal and neurodegeneration models and in ageing studies – often as a tool compound to test whether stabilising cardiolipin changes readouts such as mitochondrial respiration or reactive oxygen species production.

Elamipretide has also been evaluated in clinical trials by its developer. The SS-31 sold here is a research reagent.

Handling, reconstitution and storage

Store the lyophilised vial at −20 °C, dry and protected from light; 2–8 °C is fine for short periods. Reconstitute with bacteriostatic water, added slowly along the vial wall, and swirl gently – don't shake. Keep the solution at 2–8 °C, use it within about four weeks and avoid repeated freeze–thaw cycles.
